The plan is to have the wagon train start in Missouri in April-ish and end in Oregon in October-ish.
What part of Missouri?

The major trails overlapped & shared routes in Kansas. The Oregon Trail headed quickly north in Nebraska. Routes varied by season as travelers needed grass for livestock & water sources.
The Santa Fe Trail "started" at different points as the population drifted West. A site in the Missouri River near Boonville is among these "starts." New Franklin has since moved up the bluff!
Independence & Westport, now parts of Kansas City, were the major departure points. Mahaffie Stage Stop in Olathe, Kansas is a documented stop still standing. Academy member Iphigenia (Laura) worked there.
Lots of good information on-line and in Kansas City for you.
Captain Marcy's book has information -- lots of period information -- about the various trails.

Ms. Jean has mentioned Independence and Westport (today's Kansas City) as starting points on the Oregon, California and Mormon Trails going west starting in the 1840s. Other departure points along the Missouri River are Fort Leavenworth, Weston, Atchison, St. Joseph (a major starting point), Old Fort Kearney/Nebraska City, and Council Bluffs (on the east side of the river, crossing to today's Omaha). The latter was also a major starting point.

Travelers had to cross the Missouri River. All the named locations could either be forded or ferried or both. No bridges at the time! Sometimes you could cross on the ice....

My wagon train is planning on starting at the official starting point and ending at the official end. As others have stated, there were as many starting points as there were pioneers, and almost as many jumping off points it seems. Independence, Missouri is the official beginning of the Oregon Trail and Oregon City is the official end. Of course the beginning of YOUR trail would be your house, and the end of YOUR trail would be where you chose to settle, but Independence was chosen for the official beginning because it was one of the first major jumping off points (other towns took over in popularity later) and Oregon City was chosen as the official end because that's where you had to go to make your land claim legal.

Oh absolutely! In Kansas City, Highway 56 is considered the Santa Fe Trail route, closest you can get. My Dad has followed the Trail westbound with his camper. Council Grove, Kansas is considered a prime site with the Mahaffie Stage Stop. You can pick from The Wet Route and The Dry Route. The end in Santa Fe is the early town square, great bakery in La Fonda, The Inn At The End of the Santa Fe Trail. Wow... So late in replying here... Just now got internet at the new house, though we have been here over a month and a half. Yes. I still want to do this. It will probably be a little postponed due to the twins, and I still need a strategy, but it has not been put in the indefinitely postponed category just yet. I got a lead yesterday that might prove useful to the cause. If I can figure out how to start it will be much easier to get going.

Hi Heidi - it's so good to hear from you. I think we all thought that you were just a tad busy with the babies and your move. Photos of you and the twins at the Fort Nisqually event were posted on the Western Region ALHFAM Facebook page by Peggy at Ft. Nisqually along with photos of the event. I would think that all of us here would like some photos of the babies in their historical outfits, and you too  . Muriel
